腫瘤壞死因子α（TNF-α）由中性粒細胞，活化的淋巴細胞，巨噬細胞，NK細胞，LAK細胞，星形膠質(zhì)細胞內(nèi)皮細胞，平滑肌細胞和一些轉(zhuǎn)化細胞產(chǎn)生。TNF-α以分泌的可溶性形式和膜錨定形式存在，兩者都具有生物活性。TNF-α的天然形式是糖基化的，但非糖基化的重組TNF-α具有可比性。據(jù)報道，TNF的生物活性天然形式 - α是一種三聚體。人和鼠TNF-？在氨基酸水平上顯示出約79%的同源性，并且兩個物種之間的交叉反應(yīng)性。已經(jīng)描述了兩種類型的TNF--α受體，幾乎所有研究的細胞類型都表明存在一種或兩種這些受體類型。
Tumor necrosis factor alpha (TNF-α) is produced by neutrophils, activated lymphocytes, macrophages, NK cells, LAK cells, astrocytes endothelial cells, smooth muscle cells and some transformed cells. TNF-α occurs as a secreted, soluble form and as a membrane-anchored form, both of which are biologically active. The naturally-occurring form of TNF-α is glycosylated, but non-glycosylated recombinant TNF-α has comparable. The biologically active native form of TNF--α is reportedly a trimer. Human and murine TNF-? show approximately 79% homology at the amino acid level and cross reactivity between the two species. Two types of receptors for TNF--α have been described and virtually all cell types studied show the presence of one or both of these receptor types.
