Noggin屬于一組可擴散蛋白質，它們與TGF-α家族的配體結合，并通過抑制它們進入信號受體來調節(jié)其活性。Noggin最初被確定為BMP-4拮抗劑，其作用對于頭部和其他背部結構的正常形成至關重要。因此，Noggin已被證明可以調節(jié)其他BMP的活性，包括BMP-2，-7，-13和-14。小鼠中Noggin的靶向缺失導致產前死亡和隱性表型顯示嚴重畸形的骨骼系統(tǒng)。相反，在成熟成骨細胞中過度表達Noggin的轉基因小鼠顯示出成骨細胞分化受損，骨形成減少和嚴重的骨質疏松癥。人類爪蟾的氨基酸序列與非洲爪蟾、大鼠和小鼠的氨基酸序列高度同源。
Noggin belongs to a group of diffusible proteins which bind to ligands of the TGF-α family and regulate their activity by inhibiting their access to signaling receptors. Noggin was originally identified as a BMP-4 antagonist whose action is critical for proper formation of the head and other dorsal structures. Consequently, Noggin has been shown to modulate the activities of other BMPs including BMP-2,-7,-13, and -14. Targeted deletion of Noggin in mice results in prenatal death and recessive phenotype displaying a severely malformed skeletal system. Conversely, transgenic mice over-expressing Noggin in mature osteoblasts display impaired osteoblastic differentiation, reduced bone formation, and severe osteoporosis. The amino acid sequence of human noggin is highly homologous to that of Xenopus, rat and mouse.
